چکیده

We used light scattering and ultracentrifugation to study the binding ratio of myosin and actin from normal and failing dog hearts and to determine the effect of temperature and adenosine triphosphate (ATP) on the binding. For comparison, similar studies were done on myosin and actin from rabbit skeletal muscle. We found a higher combining ratio, by weight, for cardiac myosin and actin (4.8 : 1) than for skeletal myosin and actin (4.4 : 1). The dissociation constants were similar at 24°C for cardiac and skeletal proteins. But at 10°C, the dissociation constant was greater for cardiac proteins, and the difference appeared more pronounced when phosphate was used to buffer the solutions. The dissociation constant at 10°C was greater for myosin and actin from failing dog hearts. ATP caused dissociation of cardiac and skeletal F-acto-heavy meromyosin and F-actomyosin. There was evidence that the binding of cardiac proteins may not be as tight as that of skeletal muscle proteins. ADDITIONAL

Differential effects of pH on calcium activation of myofilaments of adult and perinatal dog hearts. Evidence for developmental differences in thin filament regulation.

Our results show that calcium activation of myofilament preparations of dog heart in the perinatal period is unaffected by a reduction in pH from 7.0 to 6.5, which, in adult heart myofilaments, induces a 0.4 pCa unit (-log molar free calcium concentration) rightward shift in the relation between pCa and myofibrillar adenosine triphosphatase activity. Acidic pH also had no effect on calcium bind...

Distribution and structure-function relationship of myosin heavy chain isoforms in the adult mouse heart.

The two cardiac myosin heavy chain isoforms, alpha and beta, exhibit distinct functional characteristics and therefore may be distributed regionally within the heart to match the functional demands of a specific region. In adult mouse hearts, which predominantly express alpha-myosin heavy chain, we observed high concentrations of beta-myosin in distinct areas such as at the tip of papillary mus...

The amino acid composition of actin and myosin and Ca2+-activated myosin adenosine triphosphatase in chronic canine congestive heart failure.

The Ca2+-activated myosin ATPase and the amino acid compositions of actin and myosin were determined for preparations from chronically failing dog hearts. Hypertrophy and congestive heart failure were produced by combined tricuspid valve insufficiency and pulmonary artery stenosis. Control, shamoperated, and noncardiac circulatory failure (inferior vena cava constriction) dogs also were studied...

Role of tropomyosin in actin filament formation in embryonic salamander heart cells

Recessive mutant gene c in Ambystoma mexicanum embryos causes a failure of the heart to function even though initial heart development appears normal. An analysis of the constituent proteins of normal and mutant hearts by SDS-poly-acrylamide gel electrophoresis shows that actin (43,000 daltons) is present in almost normal amounts, while myosin heavy chain (200,000 daltons) is somewhat reduced i...
